Upon contact with bacterial cell wall structures (typically LPS, lipopolysaccharides) IL-1b and IL-6 cytokines are intensely produced by endothelial cells. Increasing amounts of IL-1b (1) affect the activity of the hypothalamus (which leads to a rise in body temperature (fever)) and (2) trigger the expression of adhesion factors on endothelial cells to enable transmigration (also called diapedesis) of immunocompetent cells, such as phagocytes, lymphocytes and others. After arrival of immune cells at the inflammatroy site, TNF-a activates macrophages and natural killer cells; IL-6 promotes cytotoxic T cells; both IL-6 and TNF-a stimulate production of acute phase reactant protein by the liver; and ensure expression of COX-2 that results in production of Prostaglandins (enhances inflammation and pain signalling).
